| 南方根结线虫Meloidogyne incognita是严重为害番茄生产的重要病原线虫。为探寻对其具有高效防控作用的生防菌株, 本研究从番茄根结线虫病发病严重的地块分离纯化获得菌株HN01, 采用形态学观察及理化特性检测, 并结合多基因序列分析等方法进行菌株鉴定。通过菌株发酵液对2龄幼虫的致死率、卵的孵化抑制率以及对线虫体内酶活性的影响等来评价该菌株对南方根结线虫的生物活性, 并结合菌株HN01对番茄植株防御酶活性的影响、温室盆栽和田间试验评估其防治效果。结果表明:菌株HN01鉴定为婴儿芽胞杆菌Bacillus infantis, 其发酵液原液处理2龄幼虫24 h和48 h后, 校正死亡率分别为94.82%和96.77%;处理48 h, 对根结线虫卵孵化抑制率为89.96%, 且处理后的根结线虫2龄幼虫体内CAT与AChE酶活性均明显降低。菌株HN01可以提高番茄叶片及根部SOD, CAT, PAL和PPO活性。温室盆栽试验中菌株HN01防效为83.08%, 田间试验中菌株HN01处理组番茄平均株高比对照组高22.06%, 田间防效为80.43%。综上所述, 婴儿芽胞杆菌HN01对南方根结线虫具有较高的生物活性, 可有效控制番茄根结线虫病的发生, 促进植株生长, 具有较大的研究与应用潜力。 |

| Meloidogyne incognita is a major pathogenic nematode severely damaging tomato production. To explore biocontrol strains with high efficacy against it, strain HN01 was isolated and purified from a field with severe tomato root-knot nematode disease. Strain identification was performed using morphological observation, physico-chemical characterization, and genetic sequence analysis. The bioactivity of this strain against M.incognita was evaluated based on the mortality of second-stage juveniles (J2), the egg hatching inhibition rate, and the effects on nematode enzyme activities by its fermentation broth. The control efficacy was further assessed by examining the influence of strain HN01 on tomato plant defense enzyme activities, along with greenhouse pot and field trials. The results indicated that strain HN01 was identified as Bacillus infantis. After 24 h and 48 h of treatment with the original fermentation broth, the corrected mortality rates of J2 were 94.82% and 96.77%, respectively. The egg hatching inhibition rate was 89.96% after 48 h of treatment. Furthermore, the activities of both CAT and AChE activities in the treated J2 were significantly decreased. Strain HN01 could enhance the activities of SOD, CAT, PAL, and PPO in tomato leaves and roots. In the greenhouse pot experiment, the control efficacy of strain HN01 reached 83.08%. In the field trial, the average plant height in the HN01 treatment group was 22.06% higher than that in the control group, and the field control efficacy was 80.43%. In conclusion, B.infantis HN01 exhibits high bioactivity against M.incognita, effectively controls tomato root-knot nematode disease, promotes plant growth, and demonstrates considerable potential for both research and application. |
