| 琯溪蜜柚为我国知名特色柚类主栽品种, 由间座壳菌Diaporthe spp.引起的黑点病是当前琯溪蜜柚生产上的主要病害之一。为明确福建省琯溪蜜柚间座壳菌种类的多样性, 本研究采集平和县13个果园健康和具有黑点病典型症状的蜜柚的叶、枝梢和果实进行组织分离纯化, 根据菌落形态特征观察和ITS序列分析, 共获得97株间座壳菌, 从中选取13株进行多基因(ITS、CAL、TEF和TUB)系统发育分析。结果表明, 其分属于长喙间座壳D.longicolla、D.fructicola、杜仲间座壳D.eucommiae、光叶子花间座壳D.glabrae、Diaporthe sp.和菜豆间座壳D.phaseolorum。蜜柚离体果实刺伤接种结果表明, D.longicolla、D.eucommiae和D.fructicola能够引起蜜柚果实出现蒂腐或脐腐症状, 其中D.longicolla为优势种。本研究证实了福建省平和县琯溪蜜柚间座壳菌的种类具有多样性, 琯溪蜜柚为D.longicolla、D.fructicola、D.glabrae、Diaporthe sp.和D.phaseolorum等5种间座壳菌的新寄主, 为间座壳菌引起的琯溪蜜柚黑点病有效防控提供了科学依据。 |

| Guanxi honey pomelo is a famous and popular Citrus species native in China. Guanxi honey pomelo melanose disease, which caused by Diaporthe spp., is one of the most destructive diseases that seriously damages pomelo production. In order to clarify the species of Diaporthe associated with Guanxi honey pomelo, healthy and melanose symptomatic leaves, fruits and twigs of Guanxi honey pomelo samples, were collected from 13 orchards in Pinghe county for tissue isolation and purification. A total of 97 Diaporthe isolates were obtained based on their colony characteristics, morphological characteristics and ITS sequence identification. Thirteen representative isolates were chosen for multi-gene phylogenetic analyses of internal transcribed spacer region (ITS), calmodulin (CAL), translation elongation factor (TEF) and beta-tubulin (TUB) sequencing data. The isolates were identified to be D.longicolla, D.fructicola, D.eucommiae, D.glabrae, Diaporthe sp. and D.phaseolorum, respectively. In vitro fruit wound inoculation assay revealed that D.longicolla, D.eucommiae and D.fructicola were able to cause stem-end or blossom-end rot on pomelo fruits, with D.longicolla being the dominant species. The results confirmed that the species diversity of Diaporthe associated with Guanxi honey pomelo in Pinghe county, Fujian province. To our knowledge, D.longicolla, D.fructicola, D.glabrae, Diaporthe sp., and D.phaseolorum were first reported to be associated with Guanxi honey pomelo. Such information would be useful for making suitable management strategies for the control of Guanxi honey pomelo melanose disease in the future. |
