      枸杞Lycium barbarum是我国西北地区重要经济林树种。在种植过程中, 枸杞白粉病是一种常发病害, 可造成严重的经济损失。茉莉酸是植物重要的信号分子, 在诱导植物抵御病原菌侵染过程中发挥关键作用, 但在枸杞抗白粉病过程中茉莉酸途径关键基因尚不明晰。本研究以白粉病抗感性差异的‘宁杞1号’和黄果枸杞为材料, 以叶片转录组测序数据为基础, 分析了茉莉酸生物合成、信号转导和代谢关键基因表达的差异, 并使用RT-qPCR技术进行验证。结果显示, 在‘宁杞1号’和黄果枸杞中, 共鉴定到了5 713个DEGs, 其中包括2 611个上调表达和3 102个下调表达的基因。GO和KEGG富集分析发现, 茉莉酸生物合成富集了10个DEGs, 茉莉酸信号转导富集了5个DEGs, 茉莉酸代谢共富集了6个DEGs。挑选了16个DEGs进行RT-qPCR验证, 发现其表达模式与转录组数据趋势一致。相较于黄果枸杞, 抗病品种‘宁杞1号’中参与茉莉酸相关防御反应、信号转导及次生代谢相关的基因表达水平显著上调。因此, 与茉莉酸途径相关的16个DEGs可能是枸杞抗白粉病过程中茉莉酸途径关键候选基因。研究结果为进一步研究枸杞白粉病的抗病机制、选育抗病品种提供了基础。
英文摘要:
      Lycium barbarum is an important economic forest tree species in northwest China. During cultivation, powdery mildew is a common disease that can cause serious economic losses. Jasmonic acid is an important signalling molecule in plants and plays a key role in inducing plant resistance to pathogen infection. However, the key genes involved in the jasmonic acid pathway during powdery mildew resistance in L.barbarum remain unclear. In this study, the powdery mildew-resistant cultivar ‘Ningqi 1’ and the susceptible Huangguogouqi were used as materials. Based on leaf transcriptome sequencing data, differences in the expression of key genes involved in jasmonic acid biosynthesis, signal transduction, and metabolism were analyzed, and the results were further verified using RT-qPCR technology. The results showed that a total of 5 713 differentially expressed genes (DEGs)were identified between ‘Ningqi 1’ and Huangguogouqi, including 2 611 up-regulated and 3 102 down-regulated genes. GO and KEGG enrichment analyses revealed that 10 DEGs were enriched in jasmonic acid biosynthesis, 5 DEGs in jasmonic acid signal transduction, and 6 DEGs in jasmonic acid metabolism. Sixteen DEGs were selected for RT-qPCR validation, and the results showed that their expression patterns were completely consistent with the transcriptome data trends. Compared with the Huangguogouqi, the expression levels of genes involved in jasmonic acid-related defence responses, signal transduction and secondary metabolism were significantly upregulated in the disease-resistant variety ‘Ningqi 1’. Therefore, these 16 DEGs related to the jasmonic acid pathway may serve as key candidate genes involved in powdery mildew resistance in L.barbarum, providing a research basis for further studies on the disease resistance mechanism of L.barbarum powdery mildew and the breeding of disease-resistant cultivars.
