| 为明确西藏青稞赤霉病病原菌组成及其致病力, 采用单孢分离和分子生物学鉴定法对2021年和2022年从西藏“一江两河”地区采集的共316份青稞样品进行镰刀菌分离鉴定, 并对占数量优势镰刀菌种进行致病力分析, 并采用高效液相色谱串联质谱法测定其接种青稞及大米培养基后毒素污染情况。结果显示:316份青稞样品中共获得8种307株镰刀菌, 包括134株燕麦镰刀菌Fusarium avenaceum、88株木贼镰刀菌F.equiseti、52株F.gamsii、13株锐顶镰刀菌F.acuminatum、12株早熟禾镰刀菌F.poae、5株柔毛镰刀菌F.flocciferum、2株层出镰刀菌F.proliferatum和1株芬芳镰刀菌F.redolens。其中, 国内首次从麦类作物中分离鉴定到F.gamsii。对数量超过10株的5个镰刀菌种进行致病力分析发现, 除F.poae外, 其他4个种均可引致青稞赤霉病。F.acuminatum和F.avenaceum侵染后病情发展曲线下面积和总镰刀菌毒素含量均显著高于F.equiseti和F.gamsii (P<0.05), 且病情发展曲线下面积与总镰刀菌毒素含量呈极显著正相关(P<0.01)。此外, 上述镰刀菌在大米培养基上的产毒种类及能力与致病力测定结果趋势一致。 |

| To clarify the composition and pathogenicity Fusarium species causing Fusarium head blight (FHB) of highland barley (qingke) in Xizang, a total of 316 freshly harvested qingke samples collected from the “One River and Two Tributaries” region in 2021 and 2022 were used for Fusarium isolation and identification by single-spore isolation and molecular biological methods. Pathogenicity analysis of the dominant Fusarium species was performed in accordance, and high-performance liquid chromatography-tandem mass spectrometry was employed to determine mycotoxin contamination in inoculated qingke and rice culture media. The results showed that 307 Fusarium isolates belonging to 8 species were obtained from the 316 qingke samples, including 134 isolates of F.avenaceum, 88 of F.equiseti, 52 of F.gamsii, 13 of F.acuminatum, 12 of F.poae, 5 of F.flocciferum, 2 of F.proliferatum, and 1 of F.redolens. Among them, F.gamsii was isolated and identified from cereal crops in China for the first time. Pathogenicity analysis of the five Fusarium species with more than 10 isolates showed that, except for F.poae, the other four species were all capable of causing FHB of qingke. The area under the disease progress curve (AUDPC) and total mycotoxin content caused by F.acuminatum and F.avenaceum were significantly higher than those caused by F.equiseti and F.gamsii (P<0.05), and AUDPC was significantly positively correlated with total mycotoxin content (P<0.01). In addition, the types and production capacity of mycotoxins produced by these Fusarium species on rice culture medium were consistent with the pathogenicity results. |
