      为明确云南省雪茄专用烟草植株表现矮化、叶片卷曲及脉突等症状的病毒病原, 自主要种植区德宏州和玉溪市采集病样, 通过PCR检测、全基因组克隆及测序, 从11份病样中获得13条菜豆金色花叶病毒属Begomovirus病毒DNA-A全序列及5条beta卫星和5条alpha卫星全序列。13条DNA-A序列比对结果显示, 均为烟草曲茎病毒(tobacco curly shoot virus, TbCSV), 但分属2个株系——DH和YX株系; beta卫星为4种, 分别为番茄曲叶病毒beta卫星(tomato leaf curl betasatellite, ToLCB)、云南辣椒曲叶病毒beta卫星(pepper leaf curl Yunnan betasatellite, PepLCYnB)、泰国番茄黄化曲叶病毒beta卫星(tomato yellow leaf curl Thailand betasatellite, TYLCTHB)及赛葵黄脉病毒beta卫星(malvastrum yellow vein betasatellite, MaYVB); alpha卫星为3种, 分别为印度胜红蓟黄脉病毒alpha卫星(ageratum yellow vein India alphasatellite, AYVInA)、烟草曲茎病毒alpha卫星(tobacco curly shoot alphasatellite, TbCSA)及云南烟草曲叶病毒alpha卫星(tobacco leaf curl Yunnan alphasatellite, TbLCYnA)。重组分析表明, TbCSV-DH是TbCSV分离物Y-SC118和中国番茄黄化曲叶病毒(tomato yellow leaf curl China virus, TYLCCNV)分离物YN1771重组产生, 而TbCSV-YX株系由分离物YN7580-1和Tb01重组产生;中性检测分析表明, TbCSV-DH和TbCSV-YX株系进化压力不同。上述研究表明, 侵染云南省雪茄专用烟草的病毒为TbCSV, 并伴随不同的beta和alpha卫星。
英文摘要:
      To identify the viral pathogens associated with cigar tobacco showing symptoms of dwarfing, leaf curling and vein enation in Yunnan province, symptomatic samples were collected from major production areas in Dehong prefecture and Yuxi city. Through PCR detection, whole-genome cloning, sequencing, a total of 13 complete DNA-A sequences of begomoviruses, five betasatellite and five alphasatellite sequences were obtained from 11 samples. Sequence analysis showed that all DNA-A sequences belonged to tobacco curly shoot virus (TbCSV), but were classified into two strains, DH and YX. Four species of betasatellites were identified, namely tomato leaf curl betasatellite (ToLCB), pepper leaf curl Yunnan betasatellite (PepLCYnB), tomato yellow leaf curl Thailand betasatellite (TYLCTHB) and malvastrum yellow vein betasatellite (MaYVB). Three species of alphasatellites were identified, namely ageratum yellow vein India alphasatellite (AYVInA), tobacco curly shoot alphasatellite (TbCSA) and tobacco leaf curl Yunnan alphasatellite (TbLCYnA). Recombination analysis indicated that TbCSV-DH likely originated from recombination between TbCSV isolate Y-SC118 and tomato yellow leaf curl China virus (TYLCCNV) isolate YN1771, whereas TbCSV-YX likely originated from recombination between isolate YN7580-1 and Tb01. Neutrality tests analysis revealed that TbCSV-DH and TbCSV-YX were subjected to different evolutionary pressures. These results demonstrate that cigar tobacco in Yunnan province is infected by TbCSV in association with diverse betasatellites and alphasatellites.
