| 重金属相关蛋白(heavy metal-associated proteins, HMPs)是一类广泛参与重金属稳态调控的蛋白, 在植物生长发育中参与离子运输、稳态维持及营养运输等生理过程。目前针对HMP家族成员的研究多集中于金属缓冲与分配功能, 其在病原胁迫下的表达模式及抗逆机制尚未见报道。本研究对从毛果杨基因组中鉴定出的HMP家族成员进行蛋白质理化性质、基因结构及启动子顺式作用元件等生物学特性的综合分析。通过RNA-Seq分析和实时荧光定量PCR对葡萄座腔菌Botryosphaeria dothidea侵染胁迫下, 杨树HMP家族成员的表达进行分析。结果表明, 从毛果杨全基因组筛选鉴定出84个PtrHMPs, 系统发育分析将其划分为3个亚家族。这些基因的功能分化主要由不同的基因结构、基序组合、26对基因的片段复制事件(Ka/Ks=0.27)以及亚家族特异性顺式调控模块所驱动。48个PtrHMPs含N-糖基化位点, 6个PtrHMPs具有信号肽, 4个PtrHMPs具有跨膜螺旋结构。跨物种共线性分析显示, PtrHMPs与大豆HMPs的同源性最高(89对同源基因), 显示出HMP家族在双子叶植物中的进化保守性。山新杨PdPapHMP3、PdPapHMP16、PdPapHMP57和PdPapHMP62对葡萄座腔菌胁迫表现出显著的诱导表达。综上所述, 本文系统地揭示了PtrHMPs的生物学特性及其在葡萄座腔菌胁迫下的表达特征。本研究的结果将为后续杨树抗病性状改良和抗病品种的遗传育种奠定基础。 |

| Heavy metal-associated proteins (HMPs) are a class of proteins widely involved in the regulation of heavy metal homeostasis. They play crucial roles in plant growth and development, participating in physiological processes such as ion transport, homeostasis maintenance, and nutrient transport. Current research on HMP family members primarily focuses on their roles in metal buffering and distribution, while their expression patterns and stresses resistance mechanisms under pathogen stress remain largely unexplored. In this study, HMP family members were identified from the genome of Populus trichocarpa. Their biological characteristics, including physicochemical properties, gene structure and cis-acting elements in promoters, were comprehensively analyzed. Expression profiling under Botryosphaeria dothidea stress was performed using RNA-Seq analysis and RT-qPCR validation. The results demonstrated that 84 PtrHMPs were identified from the genome and phylogenetically classified into three subfamilies. Functional divergence among these genes was primarily driven by variations in gene structure, motif composition, 26 segmental duplication events (Ka/Ks=0.27), and subfamily-specific cis-regulatory modules. Among them, 48 PtrHMPs contained N-glycosylation sites, six possessed signal peptides, and four contained transmembrane helices. Cross-species collinearity analysis revealed that PtrHMPs had the highest homology with Glycine max (89 homologous gene pairs), indicating evolutionary conservation of the HMP family in dicotyledonous plants. Furthermore, PdPapHMP3, PdPapHMP16, PdPapHMP57, and PdPapHMP62 in P.davidiana×P.alba var. pyramidalis were significantly induced under B.dothidea stress. Collectively, this work systematically elucidates the biological features of PtrHMPs and their expression patterns under B.dothidea challenge, providing a theoretical basis for improving disease-resistant traits and breeding resistant poplar varieties. |
