      番茄黄化曲叶病毒(tomato yellow leaf curl virus, TYLCV)是一种全球性分布的植物病毒, 严重为害番茄、烟草等作物。为探究其致病机制, 本研究以TYLCV编码的运动蛋白V2为诱饵, 从本氏烟中筛选并鉴定出与其互作的肌球蛋白NbMyosin Ⅺ-2(GenBank: DQ875135.1)。通过双分子荧光互补、共定位及免疫共沉淀试验证实:V2与NbMyosin Ⅺ-2直接互作, 且二者在微丝上形成点状复合体。基因沉默试验表明, 沉默NbMyosin Ⅺ-2会抑制TYLCV对本氏烟的侵染, 阻碍TYLCV运动复合物V2-C5向胞间连丝的定位, 导致其滞留于细胞质, 但不影响V2的沉默抑制子功能。此外, 瞬时过表达NbMyosin Ⅺ-2显性负突变体的植株可显著抑制TYLCV、烟草曲茎病毒及云南番茄曲叶病毒的侵染。本研究揭示了NbMyosin Ⅺ-2通过与V2互作调控双生病毒的细胞间运动的分子机制, 为抗病毒策略的研发提供了新靶点。
英文摘要:
      Tomato yellow leaf curl virus (TYLCV) is a globally distributed plant virus that causes severe damage to crops such as tomato and tobacco plants. To investigate its pathogenic mechanisms, the TYLCV-encoded movement protein V2 was used as bait to screen and identify interacting protein from Nicotiana benthamiana. A myosin motor protein, NbMyosin Ⅺ-2 (GenBank: DQ875135.1), was identified as a V2-interacting partner. Bimolecular fluorescence complementation, co-localization, and co-immunoprecipitation assays confirmed that V2 directly interacts with NbMyosin Ⅺ-2, and that they form punctate complexes along actin filaments. Gene silencing assays revealed that knockdown of NbMyosin Ⅺ-2 inhibited TYLCV infection in N.benthamiana and disrupted the localization of the TYLCV movement complex V2-C5 to plasmodesmata, trapping it in the cytoplasm, without affecting the silencing suppressor activity of V2. Furthermore, transient overexpression of a dominant-negative mutant of NbMyosin Ⅺ-2 significantly inhibited infection by TYLCV, tobacco curly shoot virus, and tomato leaf curl Yunnan virus. This study indicates that NbMyosin Ⅺ-2 regulates the intercellular movement of geminiviruses through interaction with V2, providing a potential target for the development of antiviral strategies.
