2023年6月, 在新疆伊犁地区新源县改良场果园种植的桤叶唐棣Amelanchier alnifolia, 观察到疑似感染梨火疫病的症状。采集疑似病组织进行室内分离、纯化, 对分离菌株进行形态特征鉴定、致病性测定、烟草过敏性反应以及特异性引物PCR扩增及序列分析。结果表明, 分离菌株的菌落形态特征与梨火疫病菌Erwinia amylovora一致, 室内桤叶唐棣叶片针刺接种结果显示, 病菌沿叶片主叶脉向四周扩散侵染, 叶柄处有菌脓产生; 烟草过敏性反应明显; 系统发育分析显示分离菌株与梨火疫病菌位于同一分支,支持率达99%。综上可确定桤叶唐棣上分离到的菌株为梨火疫病菌。此为桤叶唐棣火疫病在中国的首次发生报道。 |

In June 2023, suspected symptoms of fire blight were observed on Amelanchier alnifolia planted in an orchard at the Gailiangchang Experimental Station in Xinyuan county, Yili prefecture, Xinjiang. Suspected diseased tissues were collected for laboratory isolation and purification. The isolates were identified based on colony morphology, pathogenicity tests, hypersensitive reaction on Nicotiana benthamiana, and PCR amplification using specific primers followed by sequence analysis. The results showed that the colony morphology of the isolates matched that of Erwinia amylovora, the causal agent of fire blight. Inoculation of A. alnifolia leaves by pinprick led to symptom development along the main leaf veins and bacterial ooze at the petiole. A clear hypersensitive response was observed in tobacco. Phylogenetic analysis revealed that the isolated strains and E. amylovora clustered in the same clade with a bootstrap support of 99%. These findings confirm that the isolated strain from A. alnifolia is E. amylovora, and this represents the first report of fire blight on A. alnifolia in China. |
