为评价从番茄潜叶蛾Tuta absoluta幼虫僵虫中分离的虫生真菌在害虫生物防治中的应用潜力, 结合形态学特征及ITS序列分析, 对分离菌株进行鉴定及生物学特性测定, 并采用浸渍法测定其对番茄潜叶蛾2~4龄幼虫的毒力。结果表明, 分离菌株为球孢白僵菌Beauveria bassiana, 命名为TaBb2405。该菌株在25℃、PDA培养基上生长良好, 菌落日生长速率为4.45 mm/d, 培养至第12天菌落单位面积产孢量为2.73×105个/mm2。随着该菌株孢子悬浮液浓度的增加, 其对番茄潜叶蛾2~4龄幼虫的毒力逐渐增强。当孢子悬浮液浓度为1.0×108个/mL时, 2~4龄幼虫7 d累计校正死亡率分别可达82.22%、80.95%和80.00%; LC50分别为6.44×104、6.21×104个/mL和1.38×105个/mL。球孢白僵菌TaBb2405对番茄潜叶蛾各龄幼虫的毒力均较好, 具进一步研究和开发生防制剂的潜力。 |

To evaluate the biocontrol potential of an entomopathogenic fungus isolated from cadavers of Tuta absoluta larvae, the strain was identified through morphological characteristics and internal transcribed spacer (ITS) sequence analysis. Its biological characteristics and virulence against the 2nd-4th instar larvae of T. absoluta were also evaluated. The strain was identified as Beauveria bassiana and designated as TaBb2405. It showed optimal growth on PDA medium at 25℃, with a colony growth rate of 4.45 mm/d. After 12 days of cultivation, the spore yield reached 2.73×105conidia/mm2. Virulence bioassays using the immersion method revealed that mortality increased with increasing conidial concentrations. At 1×108 conidia/mL, the 7-day cumulative adjusted mortality rates of the 2nd-4th instar larvae of T.absoluta were 82.22%, 80.95% and 80.00%, respectively. Corresponding LC50 values were 6.44×104, 6.21×104 and 1.38×105 conidia/mL. These results indicate that TaBb2405 possesses strong virulence across larval instars and holds promise for development as a biocontrol agent against T. absoluta. |
