      为探索斜纹夜蛾Spodoptera litura bHLH转录因子在斜纹夜蛾响应微生物农药胁迫过程中的作用, 采用酵母双杂交技术, 以SlbHLH为诱饵蛋白, 筛选经微生物农药处理后的斜纹夜蛾cDNA文库。经测序和BLAST比对分析, 初步筛选出38个与SlbHLH互作的蛋白。通过酵母双杂交点对点试验, 验证了候选互作蛋白SlCDA、SlCHIT、SlFCP、SlHEMO、SlLCP、SlTRYP与SlbHLH之间的互作关系。经金龟子绿僵菌、短稳杆菌和苏云金芽胞杆菌处理后的RT-qPCR结果表明, 在一定微生物农药及其配套处理模式下, SlbHLH的表达趋势与SlCHIT、SlFCP、SlHEMO、SlLCP的表达趋势一致, 与SlTRYP的表达趋势相反, 预示互作蛋白与SlbHLH可能共同参与调控斜纹夜蛾对微生物农药胁迫的响应过程。本研究为解析斜纹夜蛾bHLH转录因子及其互作蛋白的生物学功能奠定了基础。
英文摘要:
      To explore the role of the bHLH transcription factor in the response of Spodoptera litura to microbial pesticide stress, yeast two-hybrid assay was carried out to screen the cDNA library constructed from microbial pesticide-treated S.litura using SlbHLH as the bait. Through sequencing and BLAST analysis, a total of 38 candidate interacting proteins were initially identified. Point-to-point yeast two-hybrid assays confirmed the interactions between SlbHLH and SlCDA, SlCHIT, SlFCP, SlHEMO, SlLCP, and SlTRYP. RT-qPCR analysis following treatment with Metarhizium anisopliae, Empedobacter brevis and Bacillus thuringiensis revealed that the expression patterns of SlbHLH were consistent with those of SlCHIT, SlFCP, SlHEMO, and SlLCP, but opposite to that of SlTRYP under specific microbial pesticide stresses and treatment conditions. These results suggest that SlbHLH and its interacting proteins may collaboratively regulate the response of S.litura to microbial pesticide stress. This study provides a foundation for further elucidating the biological functions of the bHLH transcription factor and its interacting proteins in S. litura.
