为明确新疆伊犁河谷地区部分小麦植株矮小、叶片黄化、根系腐烂同时存在疑似孢囊线虫侵染引起的乱麻状须根团等症状的病因, 通过改良贝曼漏斗法和直接解剖根系的方法分离小麦根际的线虫, 通过光学显微镜对线虫进行形态鉴定, 并对采集的植物线虫进行28S rDNA D2-D3区和rDNA-ITS区序列扩增。结果表明, 从小麦根际分离出2种植物线虫, 分别与落选短体线虫Pratylenchus neglectus和禾谷孢囊线虫Heterodera avenae的形态特征最为相似;28S rDNA D2-D3区和rDNA-ITS区的序列与落选短体线虫和禾谷孢囊线虫的相似性分别大于99.1%和99.5%;基于28S rDNA D2-D3区和rDNA-ITS区序列构建的进化树表明, 供试2种植物线虫分别与GenBank数据库中落选短体线虫和禾谷孢囊线虫处于同一分支。研究结果表明,在小麦根际分离到的2种植物线虫分别为落选短体线虫和禾谷孢囊线虫, 这也是首次在新疆伊犁河谷地区小麦上发现短体线虫与孢囊线虫的复合侵染, 本研究结果为小麦根际线虫病害的识别与防控提供了科学依据。 |

In order to identify the causes of stunted growth of wheat plants, leaf yellowing, root rot and matted fibrous root mass suspected to be caused by cyst nematode infection in Ili Valley, Xinjiang, the nematodes in wheat rhizosphere were isolated by Baermann funnel method and direct root dissection method. Morphological identification was conducted using light microscope, and the 28S rDNA D2-D3 region and rDNA-ITS region were amplified from the collected nematodes. The results showed that there were two different nematodes in the rhizosphere of wheat, which were most similar with Pratylenchus neglectus and Heterodera avenae in morphological characteristics, respectively. The similarity of D2-D3 region and ITS region to P.neglectus and H.avenae was greater than 99.1% and 99.5%, respectively. The phylogenetic tree constructed based on D2-D3 region and ITS region sequences showed that the two studied nematodes were in the same clade as P.neglectus and H.avenae, respectively. The results showed that the two plant nematodes isolated from the wheat rhizosphere were P.neglectus and H.avenae, respectively. It was the first time that the complex infection of root-lesion nematodes and cyst nematodes was found in wheat in Ili Valley, Xinjiang. The results of this study provided a scientific basis for the identification and control of wheat rhizosphere nematodes. |
