为了探究不同枸杞品种对棉蚜Aphids gossypii Glover的化学防御响应机制, 本试验采用人工接虫法处理4个枸杞品种‘宁杞1号’(高抗虫)、‘宁杞5号’(抗虫)、‘宁杞7号’(抗虫)和‘宁杞10号’(高感虫)叶片, 接虫后0、24、48、72、96 h使用分光光度计法、比色法和羟胺法测定叶片内次生代谢物(单宁、总酚)含量、保护酶(SOD、PPO、POD)活性和营养物质(可溶性糖、氨基酸)含量。结果表明, 接蚜前后, 枸杞叶片中次生代谢物质含量、保护酶活性在4个枸杞品种中都存在显著差异, 4个品种对蚜虫取食呈现出一致的变化规律, 其中高抗品种‘宁杞1号’中次生代谢物质含量和保护酶活性最高, 高感品种‘宁杞10号’中最低; 营养物质含量在不同枸杞品种间也存在显著性差异, 总体含量仍然呈现出先增长后降低的变化趋势, 但在高感品种‘宁杞10号’中含量最高, 高抗品种‘宁杞1号’中含量最低。本试验分析了棉蚜为害对4个品种枸杞叶片内次生代谢物含量、保护酶活性、营养物质含量的影响, 为枸杞上棉蚜诱导抗性机理的研究提供理论依据。 |

To investigate the chemical defense mechanisms of different Lycium barbarum varieties in response to feeding by Aphids gossypii Glover, four wolfberry varieties, ‘Ningqi 1’ (high insect resistant), ‘Ningqi 5’ (insect resistant), ‘ Ningqi 7’ (insect resistant) and ‘ Ningqi 10’ (high susceptible) were inoculated with aphids. The changes in secondary metabolites (tannin, total phenol), protective enzyme activities (SOD, PPO, POD), and nutrient content (soluble sugars, amino acids) in leaves were examined 0, 24, 48, 72 h and 96 h after inoculation using spectrophotometry, colorimetric and hydroxylamine methods. The results showed that there were significant differences in the contents of secondary metabolites and protective enzyme activities in the four L.barbarum varieties before and after aphid infestation. The change trends of nutrient content observed generally showed an initial increase followed by a decrease, with the highest levels and activity found in the highly resistant variety ‘Ningqi 1’ and the lowest in the highly susceptible variety ‘Ningqi 10’. While, nutrient content varied significantly among the varieties. Nutrient levels showed an initial increase followed by a decrease, with the highest content observed in the highly susceptible variety ‘Ningqi 10’ and the lowest in the highly resistant variety ‘Ningqi 1’. This study analyzed the effects of aphid feeding on secondary metabolite content, protective enzyme activity and nutrient content in the leaves of four L.barbarum varieties. The findings provide a theoretical basis for further research into the resistance mechanisms induced by cotton aphid. |
