对云南省西双版纳州和昆明市东川区罹病的香蕉假茎象甲Odoiporus longicollis进行了病原分离。通过形态学鉴定及rDNA-ITS序列比对, 确定分离到的4个菌株均为苏格兰白僵菌Beauveria caledonica。通过对这4个分离株的离体培养条件探索及毒力测定, 表明离体培养的最佳条件为光周期L∥D=24 h∥0 h、温度25℃、在加有虫粉的PDA培养基上培养18 d, 此条件下的4个菌株菌落直径和产孢量最高, 其中Cs-1达到(65.17±0.74) mm和1.01×108个/cm2, 乳糖和牛肉膏分别为最适碳、氮源。生物测定结果表明, Cs-1菌株在接种浓度为1×109个/mL时对香蕉假茎象甲致死中时(LT50)最短, 为9.254 d, 致死中浓度(LC50)为3.618×106个/mL, 20 d后的致死率为86.67%。本研究首次在云南蕉园发现并鉴定的4株昆虫病原真菌均为苏格兰白僵菌, Cs-1菌株对香蕉假茎象甲具有较好的生防潜力, 可望今后应用于香蕉假茎象甲的生物防治。 |

Four fungal strains were isolated from infected Odoiporus longicollis collected in Jinghong city, Xishuangbanna prefecture and Dongchuan district in Kunming city, Yunnan province. They were identified as Beauveria caledonica based on morphological characteristics and rDNA-ITS sequence analysis. The biological characteristics of the strains under different conditions (culture temperature, light duration, medium composition, pH, carbon and nitrogen sources) and their pathogenicity to O. longicollis were investigated. The results showed that 25℃, L∥D=24 h∥0 h, 18 days of culture on PDA medium supplemented with insect meal were optimal cultured condition with the maximum colony diameter and spore production of (65.17±0.74) mm and 1.01×108 spores/cm2, respectively, for Cs-1. The most suitable carbon and nitrogen sources were lactose and beef paste. Bioassays revealed that the Cs-1 strain, at an inoculation concentration of 1×109 spores/mL, achieved an LT50 of 9.254 days, an LC50 of 3.618×106 spores/mL, and a cumulative mortality rate of 86.67% after 20 days. This study marks the first identification of B. caledonica strains in banana plantations in Yunnan. The Cs-1 strain demonstrates significant potential as a biocontrol agent against O. longicollis and warrants further investigation for field application. |
