      ‘粤硕菩提’是高产优质的无患子无性系新品种, 而无患子叶斑病的普遍发生会威胁无患子的良种化推广。本研究采用组织分离法分离病原菌, 对分离菌株进行致病性测定和形态学观察,并应用ITS、LSU、β-tub、rbp2基因序列对其进行分子生物学鉴定, 同时对病原菌的生物学特性进行测定。结果表明, 分离得到的3个代表菌株YS2-8、YS2-9、YS2-13均为Didymella segeticola, 是无患子叶斑病的新记录病原菌。病原菌生长最适宜的培养基为PCA, 最适宜生长温度为20~25℃, pH为7、连续光照条件利于其生长, 病原菌菌丝致死温度为55℃。本研究结果为深入研究无患子‘粤硕菩提’叶斑病发生流行规律及后续科学防控奠定基础。
英文摘要:
      The sapindus cultivar ‘Yue Shuo Bodhi’ represents an elite germplasm characterized by high yield and exceptional quality. However, the widespread occurrence of leaf spot disease in sapindus poses a potential threat to the propagation of improved varieties of sapindus. To clarify the pathogen responsible for the newly emerging leaf spot disease in the sapindus cultivar ‘Yue Shuo Bodhi’ and elucidate its biological characteristics, this study employed the tissue isolation method to isolate the pathogen. Morphological observations, pathogenicity assay, combined with multi-locus molecular biological identification using ITS, LSU, β-tub, and rbp2 gene sequences, were conducted to identify the pathogen. Additionally, the biological characteristics of the pathogen were investigated. The results showed that the three isolated representative strains, YS2-8, YS2-9, and YS2-13, were identified as Didymella segeticola, representing the first report of this pathogen causing sapindus leaf spot disease. PCA was found to be the most conducive medium for the mycelial growth of D. segeticola, with an optimal growth temperature ranging from 20 to 25℃. The pathogen demonstrated optimal growth at pH 7, and continuous light conditions favored its growth. The lethal temperature for the mycelium was determined to be 55℃. These findings provide a foundation for further epidemiological studies on leaf spot disease in the sapindus cultivar ‘Yue Shuo Bodhi’ and for the development of scientific control measures.
