2016年-2022年, 采集云南省临沧市永德县主要芒果种植基地疑似枯萎病样品60份, 通过对疑似样品的分离?纯化, 结合形态学特征?多基因系统发育分析及柯赫氏法则验证, 对8株代表性分离物(MG-1-1, MG-1-3, MG-1-7, MG-1-10, MG-2-1, MG-2-4, MG-2-8, MG-2-10)进行鉴定?结果表明: 8株分离物在MYEA培养基上散发出果香味, 形成大量子囊壳和子囊孢子团, 子囊孢子帽盔状; 序列同源性分析显示:分离物的βT1?RPBII?MS204和FG1093序列与长喙壳属真菌Ceratocystis manginecans菌株序列相似性为100%, 4个基因片段串联构建系统发育树显示, 8株分离物与源自阿曼和印度尼西亚的C.manginecans聚为一支, ML和BI分析支持率分别为93和0.98?选取2个分离物(MG-1-10和MG-2-8)进行致病性测定, 均能引起芒果幼苗发病且症状与田间相似?综上, 本研究明确了引起云南芒果枯萎病的病原菌为Ceratocystis manginecans? |

From 2016 to 2022, sixty suspected mango wilt disease samples were collected from the main mango cultivation base in Yongde county, Lincang city, Yunnan province. Using pathogen isolation, purification, morphological characterization, multi-gene phylogenetic analysis, and Koch’s postulates verification, eight representative isolates (MG-1-1, MG-1-3, MG-1-7, MG-1-10, MG-2-1, MG-2-4, MG-2-8 and MG-2-10) were identified. All isolates emitted a fruity aroma on MYEA medium, formed prominent perithecia and ascospore masses, and displayed hat-shaped ascospores. Sequence homology analysis revealed 100% similarity in βT1, RPBII, MS204 and FG1093 gene sequences between isolates and Ceratocystis manginecans strains from Oman and Indonesia. A phylogenetic tree constructed from these gene sequences grouped the eight isolates with C. manginecans with a bootstrap support of 93 and a Bayesian support value of 0.98. Pathogenicity tests on mango seedlings using isolates MG-1-10 and MG-2-8 revealed that they caused symptoms consistent with those observed in the field. This study thus confirms C. manginecans as the pathogen responsible for mango wilt disease in Yunnan. |
