      为明确青海草原毛虫Gynaephora qinghaiensis雌蛾性腺气味受体(odorant receptors, ORs)基因的分子特性及组织表达水平, 进而为后续基因功能的研究奠定基础, 本研究利用生物信息学方法及RT-qPCR技术, 对性腺ORs基因进行鉴定及表达谱分析?从性腺转录组数据库共筛选鉴定出13个气味受体基因(包括12个GqinORs, 1个GqinORco), 其中只有GqinOR1具有1个信号肽; 除GqinOR3?GqinOR7和GqinOR9以外, 其余GqinORs具有1~3个跨膜结构域, GqinORco具有8个跨膜结构; GqinORs二级结构以α螺旋为主?系统进化分析表明, GqinORs可能与桃蛀螟Conogethes punctiferalis和小线角木蠹蛾Streltzoviella insularis的ORs具有共同祖先?基因表达谱分析发现, 性腺ORs基因在雌雄不同组织表现出不同的表达水平(P<0.05), 13个气味受本基因在性腺中都有表达, 且GqinOR8?GqinOR10在触角和性腺中高表达?本研究为进一步研究青海草原毛虫气味受体嗅觉功能提供重要依据?

      To clarify the molecular characteristics and tissue-specific expression levels of odorant receptor (OR) genes in the sex pheromone glands of female Gynaephora qinghaiensis, this study employed bioinformatics and RT-qPCR techniques for gene identification and expression profile analysis. A total of 13 OR genes were screened and identified from the pheromone gland transcriptome database, including 12 GqinORs and 1 GqinORco. Only GqinOR1 had a signal peptide. Most GqinORs, except GqinOR3, GqinOR7 and GqinOR9, had one to three transmembrane domains, while GqinORco had eight transmembrane domains. The secondary structure of GqinORs is mainly composed of α helices. Phylogenetic analysis showed that GqinORs may share a common ancestor with ORs from Conogethes punctiferalis, and Streltzoviella insularis. Gene expression profile analysis showed that the expression levels of OR genes in different male and female tissues were significantly different (P<0.05), with all 13 target genes expressed in sex pheromone glands, and GqinOR8 and GqinOR10 were highly expressed in the antennae and sex pheromone glands. This study provides an important basis for further research on olfactory functions in the odorant receptors of G. qinghaiensis.
