      2023年6月发现,在江西省南昌市江西农业大学植物园种植的鸢尾普遍发生链格孢叶斑病。为明确该病害病原菌种类,对发病叶片进行真菌分离,对分离菌株进行致病性测定,共获得24株链格孢,在人工接种试验中均具有致病性;选择3株代表性菌株JXYW1、JXYW2、JXYW3进行培养性状和形态特征观察及多基因(rDNA-ITS、EF-1、GAPDH、Alt α 1)序列分析,这3株菌株培养性状和形态特征相同,符合文献中对鸢尾生链格孢Alternaria iridicola的描述,3株菌株间rDNA-ITS、EF-1、GAPDH、Alt α 1基因序列也完全相同,与GenBank中鸢尾生链格孢对应基因序列的相似性为100%,在多基因发育上与鸢尾生链格孢菌株YZU聚于同一个分支。据此,将江西农业大学植物园内鸢尾链格孢叶斑病病原菌鉴定为鸢尾生链格孢。
英文摘要:
      In June 2023, Alternaria leaf spot occurred commonly on iris planted in botanical garden of Jiangxi Agricultural University located in Nanchang city, Jiangxi province. In order to clarify the pathogenic fungus causing the disease, fungal isolation of the diseased leaves was conducted and pathogenicity of the isolated strains was tested. A total of 24 strains of Alternaria were obtained, all of which were pathogenic in artificial inoculation experiments. Three representative strains, JXYW1, JXYW2 and JXYW3, were selected for observation of cultural and morphological characteristics, as well as multi gene (rDNA-ITS, EF-1, GAPDH, and Alt α 1) sequence analysis. These three strains have the same cultural and morphological characteristics, which are consistent with the description of Alternaria iridicola in the literature. Sequences of rDNA-ITS, EF-1, GAPDH and Alt α 1 gene between the three strains are completely identical and share 100% similarity with the corresponding gene sequence of A.iridicola in GenBank. They are grouped together in the same branch with strain YZU of A.iridicola in multi-gene phylogenetic tree. Based on the results, the pathogen causing Alternaria leaf spot on iris in the botanical garden of Jiangxi Agricultural University was identified as A.iridicola.
