      中黑盲蝽 Adelphocoris suturalis 是一种重要的农业害虫, 主要为害棉花、果树和牧草等作物。昆虫保幼激素(juvenile hormone, JH)与蜕皮激素(molting hormone, 20E)是调控昆虫生殖的主要因素。本研究运用基因克隆、基因沉默(RNAi)、实时荧光定量PCR(qPCR)等技术研究蜕皮激素受体(ecdysone receptor, EcR)、超气门蛋白(ultraspiracle protein, USP)在中黑盲蝽生殖调控中的作用。结果表明, 中黑盲蝽 EcR 基因的ORF全长1 413 bp, 编码470个氨基酸。预测蛋白质分子量为53.65 kD, pI值为7.79。中黑盲蝽 USP 基因ORF全长1 209 bp, 编码402个氨基酸。预测蛋白质分子量为44.99 kD, pI值为7.94。与对照相比, 注射dsEcR、dsUSP后6~18 d的中黑盲蝽体内 EcR 基因、USP 基因在转录水平分别被显著抑制; 卵巢的挂卵量分别仅有对照组的31.5%、86.6%; 单雌产卵量减少36.0%~80.4%, 显著低于对照。 EcR和USP 基因沉默后,产卵前期与对照相比无显著差异; 雌成虫寿命比对照组减少2.4~5 d。上述研究表明, 沉默蜕皮激素信号通路的2个关键基因 EcR 和 USP 对中黑盲蝽的生殖力产生显著的影响, 蜕皮激素信号通路可能是中黑盲蝽生殖调控的关键信号通路之一。
英文摘要:
      Adelphocoris suturalis is an important agricultural pest, mainly damaging cotton, fruit trees and pasture. Insect juvenile hormone (JH) and molting hormone (20E) are the main factors that regulate insect fertility. In this study, gene cloning, gene silencing (RNAi), real-time quantitative PCR (qPCR) were used to clarify the roles of ecdysone receptor (EcR) and ultraspiracle protein (USP) in the reproductive regulation of A. suturalis. The ORF sequence of EcR gene was 1 413 bp, encoding 470 amino acids. The predicted molecular weight of the corresponding protein was 53.65 kD, with a pI value of 7.79. The ORF sequence of USP gene was 1 209 bp, encoding 402 amino acids. The predicted molecular weight of the corresponding protein was 44.99 kD, with a pI value of 7.94. Compared with the control, EcR gene and USP gene were significantly inhibited at the transcriptional level 6-18 days after injection of dsEcR and dsUSP. The number of eggs in the ovaries was only 31.5% and 86.6% of that in the control group, respectively; the number of eggs laid per female significantly decreased by 36.0%-80.4%, compared with the control. The pre-oviposition period of adults between the dsRNA injection and control group showed no significant difference, but the lifespan of female adults in dsRNA injection group was 2.4-5 days shorter than that of the control group. These data suggest that silencing the two genes EcR and USP of the ecdysone signaling pathway has a significant impact on the fecundity of A. suturalis, and the ecdysone signaling pathway may be one of the key signaling pathways in the reproductive regulation of A. suturalis .
