      以我国高寒地区分离、筛选出的高致病力昆虫病原线虫嗜菌异小杆线虫(Heterorhabditis bacteriophora HBN)为研究对象, 对其冷冻储存条件进行优化。结果显示, 介质中不同含水量对线虫HBN的冷冻存活率有着显著的影响; 其中含水量为50%的蛭石中线虫回收率最高, 为87.4%; 冷冻储存后的线虫经4 ℃ 24 h+室温1 h解冻利于线虫冷冻休眠后复苏; 线虫经阶段性降温较直接降温至-4 ℃的冷冻存活率提高31.6百分点; 储存介质蛭石中采用灭菌自来水较适于HBN线虫在-4 ℃冷冻存活; 7 d内冷冻储存增强了HBN线虫对寄主昆虫的侵入能力。

      One high virulence strain of EPNs, Heterorhabditis bacteriophora HBN (HBN) with cold tolerance isolated from Harbin City was used in the laboratory in this study. Storage substance species and moisture, cooling rate, thawing ways and osmotic pressure of solution in the substances were optimized. The results showed that freezing survival of HBN were significantly affected by the substances with different water contents. Recycling coefficient of H. bacteriophora HBN in the vermiculite with water content of 50% was the highest (87.4%). The way of freezing HBN at room temperature for 1h following 4 ℃ for 24h was in favor of thawing of freezing dormancy. Freezing survival rate of HBN by phasic cooling pattern was increased by 31.6 percent than by direct cooling to -4 ℃. Sterilized artificial tap water in vermiculite was conducive to anhydrobiosis of HBN. The ability of HBN infecting Galleria mellonella was enhanced in the short term freezing storage in seven days.
