为探索应用EST-ILPs分子标记技术快速鉴定菟丝子种子的方法,从网站(http:∥ibi.zju.edu.cn/pgl/pip/index.html)上公布的ILPs标记中选取旋花科番薯属(Ipomoea)已经设计好的引物10对,对菟丝子样品进行PCR扩增,寻找种间特异性条带。对菟丝子的5个近似种进行ILPs-PCR 反应结果表明:应用引物IP9进行扩增反应,恩氏菟丝子在250 bp左右具有一条特异性条带;应用引物IP4进行扩增反应,日本菟丝子在240 bp左右显示一条特征条带;通过菟丝子内含子基因型的多态性特征可以区分供试的5种菟丝子。EST-ILPs分子标记检测菟丝子近似种作为菟丝子种间鉴定的一种新的研究途径,为口岸杂草检疫鉴定提供了一种快速、准确的检测方法和科学依据。 |

To explore the method for rapid identification of Cuscuta seeds by EST-ILPs, 10 pairs of primers for Ipomoea(Convolvulaceae) were selected from the website (http:∥ibi.zju.edu.cn/pgl/pip/index.html) to amplify Cuscuta samples by PCR to find the specific band. The results of ILPs PCR of 5 related species in Cuscuta showed that Cuscuta engelmanii had a specific band of around 250 bp by using the primer IP9, and C. japonica had a specific band of around 240 bp by using the primer IP4. The polymorphic characters of the intron genotypes of Cuscuta could distinguish the 5 Cuscuta species. Using ILPs markers to identify the related species in Cuscuta is a new method for species classification in Cuscuta, which provides a new scientific evidence and identification method for quarantine. |
