      本研究探讨了UP PCR技术在玉米丝黑穗病菌遗传多样性分析中的利用可行性。在13个通用引物中,筛选出9个扩增多态性好且稳定的通用引物,共扩增出113条DNA条带,大小分布于250~2 000 bp之间,其中多态性条带为95条,为总条带数的84.07%。遗传距离为0.76处时,所有丝黑穗病菌菌株被聚为6个组。利用UP-PCR技术,可以充分展现玉米丝黑穗病菌菌株间的亲缘关系及差异性,可用于玉米丝黑穗病菌遗传多样性研究,为有效地开展玉米丝黑穗病菌的遗传进化甚至探讨病菌致病性的生理分化提供了一个新的技术方法。
英文摘要:
      Genetic diversity of Sporisorium reilianum isolates on maize collected from different geographic areas in China were analyzed by UP-PCR (universally primed PCR). The results showed that a total of 113 bands were amplified using 9 selected primers from 13 universal primers. The length of these bands ranged between 250-2 000 bp, and 95 of the 113 bands were polymorphic, accounting for 84.07% of the total. For genetic distance of 0.76, all isolates were clustered as six distinct groups. UP-PCR analysis uncovered the phylogenetic relationships and difference of the tested isolates of S. reilianum, which provided a new method for analyzing the genetic evolution and the physiological differentiation of S. reilianum.
