本试验对小麦胚芽鞘体细胞染色体制片技术进行了初步探索。结果表明,利用小麦胚芽鞘细胞可以获得理想的染色体制片,而且具有切取胚芽鞘对小麦个体植株伤害小,胚芽鞘体积大、分裂相多,易于制片,镜下观察染色体较大,杂质少,背景清晰等特点,可满足染色体核型分析和染色体分带的技术要求。初步建立起以小麦胚芽鞘为取样对象的体细胞染色体制片方法。 |

A technique for chromosome preparation using wheat coleoptile cells was introduced in this paper.The results showed that the coleoptile cell was fit for chromosome preparation and a good effect could be achieved.The characteristics of the technique were as followed: less damage to wheat individual plant, prone to production with bigger volume and more cycle phases, easier to observe under microscope, less impurity and clearer background.This technique can suffice the demands of karyotype analysis and chromosome banding.The chromosome preparation procedures with wheat coleoptile cells as sampling objects were preliminarily established. |
